The silence at the end of American Sniper.
Not only did Viggo Mortensen deflect that very real knife in real time with his sword (not CG) , he also acted through the pain in another scene when he broke his toe outside of Fangorn forest.

Endgame portal scene and Cap fighting Thanos
Speaking of Marvel I’ve never heard a theater so silenced leaving a theater like I saw after Infinity War.

Going waaaay back… Superman II.
People were going apeshit when Superman turned the table on Zod at the end. People were ready to riot over Zod. It was crazy.

hardest I’ve ever laughed at a movie.
People in the theater were laughing at how hard I was laughing. I kept losing my breath.

The female avengers girl-power moment in End Game. My whole theater jumped to their feet and the biracial, non-binary toddler that had been crying the whole movie suddenly stopped, turned to their moms and said “I don’t care what that curmudgeon Scorsese says, THIS is cinema.”

Opening show at midnight for Star Wars Phantom Menace. It was jam packed, people all over in costumes, and when the opening credits rolled the place erupted like they were at a football game.
But then we had to watch the actual movie.
Edit: I see that others have mentioned something similar as well and it looks like they were in different locations. It seems like it was the same everywhere.
Part of it was people waited in line for literally days to get tickets to that midnight showing.

The crowd being somber at the end of Saving Private Ryan even though it was a great movie. It was like walking out of a funeral having lost someone close. No-one talked as we all walked out
This one was memorable as well. I was at an advanced showing where they invited a few WWII vets and their families to screen it (and also other military personnel). 3 of the WWII vets got up and durning the opening scene at Normandy and left. The theater then stopped the movie. 1 of the 3 came back and watched the rest movie.

Dumb and Dumber. I have never seen a theater just dying with laughter like I saw during that movie back then
This and Something About Mary
My dad took me to see D&D and during the toilet scene I thought he was going to have a heart attack.
I took a date to Something about Mary and the hair scene people lost their shite

The final battle in endgame when everyone shows back up I’m not a marvel nerd but that was cool and the theater was absolutely hyped up as it was unfolding
This was bigger for my audience. We all knew but getting to see it was wild.
I’ll add walking out of Passion of the Christ with my friends from church was weird. We didn’t talk to each other. We all just walked away.

The surprise reveal in The Crying Game.
Why was anyone surprised by this? No one could tell he was a damn man?
All the reviews said it had a surprise twist that you would not see coming. So I was watching closely for what it might be and saw it coming. Same for Sixth Sense.
I likely would have been surprised by both, but telling me ahead of time there was a surprise or twist essentially spoiled them.

When Michael Gross and Reba McIntyre back into the frame with the wall of firearms in Tremors. Essen 6 went batshit.

The scene in hereditary with the daughter in the car. I think the whole theater was like wait, what?

The night “Army of Darkness” opened. Theater was packed. Some friends and I went thinking we would be the only dorks there. We were wrong. We liked the first two Evil Dead films but thought no one else did.
The first line of the movie is “My name is Ash, and I am a slave.” As soon as he said that line the entire theater yelled and cheered. After every sarcastic one liner spoken by Ash the theater reacted the same way. Loud cheers like those you hear when a walk off grand slam is hit. We were not expecting this. We went to see it again a week later and the theater was empty.

Two old men physically upset watching the opening scene of Saving Private Ryan.

Endgame when Cap pulled Mjolnir to him. Then when Spidey specifically swung into the reunion frame. Then when Cap finally said, "Avengers... Assemble."
This for me.
Although a very close second would be Revenge of the Sith, when Anakin has the Vader mask going on him the first time. The theatre was eerily silent, could hear a pin drop. I just distinctly remember the silence. The music in the movie stopped right as it was going on him, and then you just heard the mechanical air lock of it, a second of pure silence, and then the iconic Darth Vader breathing.
