Here's another, one of the biggest officiating mistakes ever
the 5th down game in 1990 , Colorado @ Missouri.
Colorado got an extra (5th) down and scored to win the game, they also won a share of the national title with Georgia Tech.

South Carolina losing to Navy in 1984.

Saints really have 3 that jump to mind. This is how I'd rank them in order:
No Call
Beastmode
Minnesota Miracle
Only change is I've always thought it was referred to as Beastquake game.

The Browns
These are all the famous ones from the 80s when they actually had good teams. There’s an entire menu of “signature losses” they’ve had since the return in 99. Basically at least one very year
Well, I'd suggest that a "signature loss" would be famous, from when a team is good

It's different than regular ineptitude. It's snatching defeat from the jaws of victory.

For LSU, I don't really count 1/9/12, because that game was never in doubt past the opening kickoff. Yeah, the defense played valiantly and kept the team in range, but the offense never showed any sign of life.
I would say basketball, I forget what year. Clarence Caesar was on the team. Jumped all over Kentucky, at one point going up 30 (70-40). Deafdome was absolutely nuts, all the way through. Tiger rappelled from the ceiling pregame, LSU totally blasted the Wildcats.
But the game wasn't over, despite LSU celebrating and clowning around to the fans. Kentucky had a comeback for the ages, and won.

Bills fans are lucky enough to have two.
Make that 3. Finally host the Chiefs in the playoffs, in a down year no less, and couldn't take advantage.

You think the Drive isn’t the signature? It launched everything for Elway.
The Broncos meltdown vs Washington has to be the loss
I think the Browns The Fumble is worse than The Drive. The win maybe the playoff game to make Ben sit there like a child?

2000 AFC divisional to ravens, 08 Div Ravens, 2021 never get over home loss to the bengals in divisional with 9 burrow sacks

I mean…
Do I even need to mention the Falcons?
This is the only answer.
That might even be the signature loss in the history of the NFL.
I can't imagine even if the NFL last for another 100+ years, that any game could top that loss. If they did a Top 10 worst playoff losses countdown on NFL Network, they'd have to call it "The Falcons choke away a 25 point lead in the 4th quarter of the Super Bowl and 9 other less bad playoff losses."

Even if they did just wide right another 40 something yard fg in the playoffs, losing a divisional round game is nowhere near as signature as a Super Bowl loss.

There's levels to it. Wide Right is definitely the signature loss for the Bills overall, but tonight was a signature loss for the McDermott-Allen regime.

Idk if this is recency bias, but does the Bills allowing Mahomes to drive and win the game with only 15 seconds left, make this list at all?

I could fill up this entire thread with NCAA Tournament college basketball games. (Phi Slamma Jamma vs. Jim Valvano, Patrick Ewing vs. Georgetown, etc.)
Also Donnie Allison and Cale Yarborough in the 1979 Daytona 500 and Roy Jones in the 1988 Olympics.

For your school, the 1998 Big 12 Championship Game. In fact, that's one of CFB's signature losses.

Idk what the Falcons signature win is (maybe 2016 NFCCG) but everyone knows the signature loss (28-3). It is undoubtedly much more well-known than any of their wins. The franchise still hasn’t recovered to be honest.
The Falcons signature win is clearly the win in Minnesota for the NFC championship.

It doesn't eclipse Wide Right as the signature loss for the Bills franchise as a whole, but it's probably the signature loss for the Allen-McDermott regime.
