There's still a major issue of the human body being accustomed to gravity.
Astronauts lose 1-2% of their bone mass per month in space. They can lose 50% of muscle mass in a few months. That's even with resistance train to help compensate.
The human body just isn't designed for low/no gravity and atrophies quickly.
The longest anyone has ever been in space in one go is 374 days, basically one year, which is nothing when it comes to longer missions and semi permanent colonies.
Until artificial gravity is a thing I think we're essentially stuck on earth.
