I’ve got to go with the opening scene of Inglourious Basterds. It’s absurd, tense, terrifying, and brilliant all at once. The stuff that’s funny at first becomes horrifying as the scene goes on and you finally figure out what Landa is actually up to. It’s a truly perfect scene: YouTube

In Dirty Harry, Harry is walking down the steps of the hospital with the wife of his wounded partner after visiting him. The wife asks Harry why he does it (police work). Harry answers, "I don't know. I really don't." That resigned sense of duty and obligation to perform a thankless job sums up the character beautifully.
Comedy:
"Cinderella story! Tears in his eyes!"
Superhero:
Batman vs Superman warehouse fight scene
Musical:
Two words - Olivia/leather

once upon a time
When pictures told the story and not talkie-talkie, conveyed the emotions, the scenario and the outcome to the audience.
Plays were all dialogue. Films were representation through pictures. Leone kept that tradition alive and did a masterful job at it. It's a Cinematic treasure.
