I've never seen Up but I always see that it's referred to in threads about tearing up or saddest movies

No one has mentioned the ending of Braveheart? Wallace's execution and the end scene got me.

Other notable mentions:
Shawshank Redemption
Castaway
Edward Scissorhands
My Girl
Pearl Harbor's ending
Gladiator's ending
John Q
Mr. Holland's Opus

I didn't want to mention this because I wanted to stick to movies like mentioned in the OP, but if we're talking TV too then my Lord yes at this. I had heard/read about this and was like "Futurama? No way" so I watched it online. I saw how much time was left in the episode close to the end and was wondering what the hell all the fuss was about. Then in that last 30-60 seconds it was just...damn.

I remember feeling like shite after I watched that episode.
I mean, I expected it to end happily...or at least normally.
But it was just miserable.
I wasn't sad; I just felt like shite.

Probably one of the reasons I stopped watching that show.

Most of mine have been mentioned.

These get me every time
UP
Braveheart
Toy Story 3
A Walk to Remember

This is a scene that just hits me like a ton of bricks. It's from a TV show, but it really really does it. Spoiler Alert for anyone who wants to watch Full Metal Alchemist.

In Casablanca, inside Rick's Cafe Americain, an obnoxious group of drunk uniformed Germans are loudly singing Nazi songs, while the rest of Rick's customers are looking glassy-eyed and depressed. Then a key character stands up and with Rick's nodded approval, tells Rick's regular band to strike up "le Marseillais" and the crowd goes wild - stands, starts singing, the Germans try to drown out the French voices and band, the French voices and the band just play louder. In the end, the Germans slink away and by this point I am in tears. "Vive le France!"
