If not already said, Peter Capaldi played as an ancient Roman in Doctor Who during the David Tenant run, then played in a 5 episode arc of Torchwood, then became the Doctor.
So he went from minor, to less minor, to the title character - within the same universe.

Keith David, who had a pretty big part as Elroy in Community, had a voice-over role in one of the earlier pillow-fight episodes.
Also, Paget Brewster had a role as an IT person in a Community episode before taking the larger role as Frankie.

Catherine Bell had a very bit part (3 lines) on the first season of JAG when it was on NBC.
When NBC killed the show and it was resurrected on CBS, she was a full time cast member.

Lucy Lawless in the "Hercules" universe.
Appeared in one of the Action Pack, pre-series Hercules movies as one character, then appeared on the series as a different character, before being cast as Xena three episodes later in the first season because Vanessa Angel was unable to play the role due to illness.
I enjoyed Xena, but always thought she was a bit too butch and had a moonface. Vanessa Angel was a babe.
Would have enjoyed this show a lot more

Edward Winter played a "Captain Halloran" on MASH before returning as Col. Flagg. Some people say this was actually Flagg's first appearance, but that he was his usual undercover self. (I believe in later appearances Flagg makes mention of this episode so that supports the theory.)

Neither is a large role, but in Seinfeld in an early season Jerry and George both date the shania Twain looking lady that they both find droning, then she also appears in an episode several seasons later as a waitress at monks (as a different character, not the ex gf)

Maybe too obvious and out of the intended purview of the thread, but I always thought the Jonah Hill in Forgetting Sarah Marshall and Get Him to the Greek two separate characters in the same universe were an unusual casting choice.

Not a featured character, but I’ve been rewatching Seinfeld and noticed the woman who plays the nazi in the limo is later Jerry’s girlfriend who refuses to eat a bite of his pie.
