Someone brought up Steve Landesberg appearing in an early episode of Barney Miller as a con-man posing as a Catholic priest... then returning as the ongoing character Sgt. Dietrich.

I'll add that Joseph Gannascoli appeared in a first season episode of the Sopranos as "Gino" the terrified bakery patron who Christopher runs off before shooting the clerk in the foot. The next time we saw him he was Vito Spatafore the gay Soprano crew captain.

Dean Charles-Chapman played Willem Lannister in Season 3 of Game of Thrones before becoming Tommen after they recast him.

Before he was Col. Potter, Harry Morgan guest starred in one episode of MASH as Major General Bartford Hamilton Steele. There are a lot of these.

He returned for a third role in the movie, playing one of the background local yokels.

John Mahoney appeared on Cheers as a jingle salesman. Then turned around and became Frasier's father on that series. (Different shows, same universe.)

Nia Long on Fresh Prince. She played the role of Claudia in one episode from Season 2 before a recurring role as Lisa later on in the show.

Peter Capaldi as well as a few a few other actors/actresses played as a one time character on Doctor Who before returning as a different role. In His case, he returned to actually be the Doctor.

On Married with Children, Ted McGinley played Peg's husband in the episode where Al's gaurdian angel (played by Sam Kinison) showed him what life would be like if he wasn't born.

Also on Golden Girls, Harold Gould appeared in an early episode as someone Rose dated but later appeared as Rose's boyfriend Miles.

S. Epatha Merkerson was in one of the early episodes of season one of Law & Order before she came back as Van Buren.

Law & Order is notorious for recycling actors.

Diane Neal was a rapist, then returned as ADA Casey Novak.

Josh Pais has played eight different characters and been on every version of L & O. He was one of the reoccurring medical examiners on the original series and was the deputy commissioner on SVU who had hard drives full of kid porn hidden throughout his house.

Lucy Lawless in the "Hercules" universe.

Appeared in one of the Action Pack, pre-series Hercules movies as one character, then appeared on the series as a different character, before being cast as Xena three episodes later in the first season because Vanessa Angel was unable to play the role due to illness.

(Fell down that hole because of a recent "Battlestar Galactica" binge.)

Crocodile Dundee 2... the bloke that gets knocked out by Donk when Nugget says “I don’t need a gun, I’ve got a Donk.” He returned in the Crocodile Dundee in Los Angeles as Mick’s cousin or buddy... that was a lazy hire.

Karen Gillian is the best example of another Dr. Who player that was actually in the Peter Capaldi guest spot I think (Pompeii ep) and returned as Amy Pond...She made it big!

Keith David was the documentary narrator on the Pillows vs Blankets episode of Community and was later Elroy in season 6.

Edit:

Thinking about it, Paget Brewster had a small role on one episode in the early seasons as well.

Reminds me that Roger Davis narrated the series opening segment on Alias Smith and Jones. After Pete Duel committed suicide, Davis took over as Duel's character, and was replaced in the opening narration by broadcaster Ralph Story.

Davis had also appeared in a Duel episode as a villain.

John Lovitz was in a couple of episodes of News Radio as different characters and then joined the regular cast as Phil Hartman's replacement after his death.

The Star Treks had a lot of these, come to think of it, especially the folks that played aliens.

Spock's father was the first Romulan on the series before returning as Sarek of Vulcan.

Tim Russ was a die-hard Trekkie and was cast in multiple small roles on Next Gen, the Generations film, and elsewhere in the ST universe before becoming Tuvok on Voyager.

Most importantly, Tim Russ was the crewman manning the giant afro-comb on Space Balls who shouts out, "WE AIN'T FOUND shite!"
