I’ve told people for years that Subway serves more mystery meat than a school cafeteria.

The SO's kid loves Subway because of her cheap arse mother who buys it when she refuses to eat her cooking. We refuse to let her get it when here. I'll buy the shite to make subs at home before I'd buy Subway again.

Heat denatures DNA. The polymerase chain reaction uses this property. Heat the DNA to about 205F or so and hold for 30 or so seconds. The two halves of the DNA double helix unzip leaving two single strands of DNA. The single strands are still viable and can still be reassembled into double stranded DNA by the proper enzymes upon a slight reduction in temperature to prevent the newly formed double strands from denaturing again. This is the backbone of the polymerase chain reaction. Mildly heat DNA to denature it, then introduce a polymerase that can withstand the temperature and then lower the temperature to allow the newly copied DNA to avoid being denatured until the copy is completed. Then turn the heat back up and repeat the cycle over and over. 1 DNA molecule becomes 2 becomes 4 becomes 8... This process is what's called "amplification".
Higher temperatures or prolonged heating, however, will physically destroy DNA, not just temporarily denature it. Canned tuna is generally cooked in a retort at temperatures well above 205F for hours on end. It wouldn't surprise me one bit if there are no viable DNA fragments left after the canning process.
