This is simply the easiest situation for anyone over eight years old. If she doesn't want to come, she can stay home. It's her husband's choice as to whether he wants to attend, also. If either one gets upset, who cares?

I can't believe any grownup needs to ask anyone what he should do. It's your party. Screw them if they have a problem.

I bet deep down coworker doesn’t want her to come because he would have more fun.

I assume she is vaccinated so what’s the issue? She is protected so if someone isn’t vaccinated it won’t affect her

Straight up ignore it, as in act like it never happened. Enjoy your birthday dinner even if she shows up and acts like a bitch. Matter of fact, if she does act like a bitch make sure she can tell that you are having a good time. Not much in life pleases me more than not letting a bitch get pleasure out of being a bitch.

B2) Tell him you're not doing that and that you understand if he won't be able to make it given the situation.

Why would you make it any more complicated than it needs to be. Unless you just really want him there because you're hoping he'll get drunk and hook up with you.
