One of my first homes was huge. . It was over 6k sq ft, had a full basement, was on 3 levels, plus the basement below. It had a 4 car detached garage with another huge room about the parking area too. That was probably another 1000 sq ft. It was massive - I'd guess close to 10k sq ft total - and had stairs everywhere. When I sold it, I sold it to some couple and the husband had just had heart surgery. Let's just say I was pretty shocked and surprised that they were the ones that bought the place.

Born in Louisiana, but grew up in California. Went back to LSU, then after college back to Southern Cal. All two story homes out there have the Master upstairs. When I moved to Texas, all the Masters are downstairs. I remember making the statement that I don't know if I'll ever get used to having the Master downstairs? Now, I wouldn't have it any other way. So, I guess you'll get used to whatever you have.

Make sure it has a dual AC system or the bedrooms upstairs will always be 5-10 degrees hotter than downstairs.

Don't do it unless you plan to stay for the long haul. Resale is much more difficult on these homes.
ETA: Let me clarify that I'm speaking for the BR area and that it may be customary in other markets. This is not a desirable trait in this market. Sorry downvoter, it's not debatable.

We do. Like it, but a dedicated HVAC is imperative. I'm early 70s and imo the exercise does me good, keeps the ol' joints flexible. 19 steps, and I make a couple extra trips a day just on principle.
Just don't take a max strength nighttime cold med unless you're already up there, lol. Been there, done that.

Three of our four BR are upstairs along with the master and laundry room. We love it.

We built basically 2 MBRs in our house. One is upstairs along with 2 kiddo BRs. Other is downstairs and could be used for taking care of an elderly parent. That one joins the rest of the house but is otherwise separate (exterior entrance, full bath, closets and walk-in shower).
We’ve basically used it as a guest room for now since all our parents are living on their own. Eventually, it will probably become our main BR, assuming we reach the age that makes stairs untenable.
As far as living in an upstairs MBR, it hasn’t presented any significant issues. It’s nice to have the whole downstairs to myself when the wife turns in early (like tonight). Also, it’s useful for keeping an eye on the kiddos, especially when they were younger. Completely separate upstairs HVAC, so that helps a bunch.

Interesting point. In the northeast, seems most people prefer to have all bedrooms on the second floor. All of our bedrooms are on the second floor and we have two sets of stairs. Our bedroom is in one direction at the top of the main stairs and the rest of the bedrooms are in the other direction.

I really like having a private balcony that can only be accessed through the master bedroom. To me that more than offsets any inconvenience.

In the Midwest and Northeast multistory houses with all bedrooms on a upper floor are the rule. Many generations of the elderly have lived out their lives climbing stairs. Health related mobility issues among the elderly are not as common as some believe, and many are only temporary.
