I wouldn't do it.
Break a a leg, tear an ACL, need hip labrum surgery, etc., not way do I want to go up and down stairs after that. Not to mention if you age it it becomes your forever home.

Biggest issue, imo. We owned one as a starter home when we first married. Entire upstairs was master bed/bath. Had no issues with temperature differences. If you have children, it can be a bit unnerving. When they’re young, you’re concerned about getting to them in case of emergency. If older, it’s a “sneak in/out at will” scenario.

Mine is upstairs...don't particularly like it but wasn't a deal breaker.
ETA:
We use the upstairs masters because we have young kids and were having to get up with them during the night. Next house the master will definitely be on main level.

My current house has the master bedroom and bathroom upstairs. One bedroom downstairs and 2 upstairs. I've never really thought about it as being a negative at all.

We have that, and like it. Things to consider:
Is there a second HVAC for the upstairs? Our house has separate units, and the upstairs one is bigger (4 ton, IIRC) because the majority of the home's LA is upstairs.
Is the laundry upstairs? Ours is, and that is a dealbreaker. I can't imagine tromping up and down with laundry. Mow the grass? Stop in the laundry room outside the MBR and strip down before taking a shower without romping through the downstairs scarring people for life.
Is the MBR above the garage? Ours is, and it has downsides. The openers are loud, and because you have a big non-climate-controlled space under you, it gets colder in the winter (I like that) and hotter in the summer. (I do not like that)
We bought this house specifically when we saw that, because we had a baby on the way. Neither of us liked the idea of a ground floor master away from the baby once they went into a crib in their own room. Then during Hurricane Harvey in 2017, we were able to move most of our furniture upstairs in case water got in (it didn't) and knew that if we had to make repairs, we could "live" upstairs during the work.
The only cons I've experienced is - if you have a health issue (wife [no pics] had a C-section and the stairs were a challenge - she lived upstairs for a week) then stairs in general are something you may not want to mess with. The electric bill is higher during the summer because heat rises and I WILL NOT sleep hot. Other than that, having a 2-story has given us a larger yard than if we had the same LA in a 1-story.

Our house in TN has all 4 bedrooms upstairs. Our rental did as well, I think it's very common around here. I don’t mind at all, but I am not constantly up and down. We also have dual zones for HVAC.
ETA: The laundry is downstairs, that part I would change if we could to be upstairs with the bedrooms.

I have and it was not an issue whatsoever. That said, I'm not elderly or infirm so my experience might have been different if I was.

I actually don’t think a two story with master upstairs sounds bad at all. I don’t like guests accessing my bedroom.
