Not sure about "the best". One of the best is New Orleans guitarist Kala Chandra. The link is a rehersal at the DBA bar on Frenchman St
Kala solo at about 2:40 (about 10 seconds of clarinet before Kala - Chloe Feoranzo also a killer musician)

Eric Clapton would be very wrong on that. He’s also gone on record saying Sonny Landreth is this best guitar player alive before too.
Marcus King is an unbelievable up and coming talent in the Bonamassa style of playing. Very aggressive, very clean, huggggge tone, incredible phrasing, vibrato, melody, fret board and theory knowledge, insane performer live, one of the best on the planet today.

Best overall guitarist over all genres is going to be one of the LA or Nashville guys, like Jack Pearson, Brent Mason, Tim Pierce, etc. Gary Clark is a average blues player, in that genre Matt schofield, Josh Smith, Arien Posien, Kirk Fletcher, and several others are at a whole other level, Clark just happened to get the record deal.

Richard Thompson is pretty damn good.

This. Gary Clark Jr. plays with a lot of feeling. He’s a very tasteful guitarist that knows when not to play. With that said, he’s nothing special as a picker. There’s probably a dozen guitarists tooling away in local bands just around Baton Rouge that are better instrumentalists than him. What Gary has is that he writes great songs, has a great voice, and has some serious stage presence. His guitar playing itself really isn’t anything uncommon.
I’d like to see Tom Morello and Gary Clark jr. duel it out to see who is the worst of the renowned “great” guitar players out there today.

Tony Rice is an exceptional talent...doesn't get much recognition because he's a bluegrass musician.

Until he either retires or dies the answer to this question is going to be Jeff Beck.

I'll go with John Mayer
I think she knows interlude guitar cover
ETA: how does he keep that steady sound that he starts off with keep going throughout the entirety of it? Even as he riffs more complicated stuff, he can hear that initial sound throughout the clip. How does he do that?

Sonny Landreth and Guthrie Trapp need to be considered.

Paul Gilbert, Buckethead and Joe Satriani are the best IMHO.

He says in the video description, "Looped through a Boomerang pedal."
