This would not be the first question I would ask in this situation, because what we first need to ascertain if they have the ability to synthesize the virus itself. While a BSL-4 lab should have this ability, the Wuhan lab was relatively new, and you should be able to trace the equipment required to synthesize a reverse genetics system for the virus. The lab there was fully opened in 2018, and the last version of Methods in Molecular Biology to have a chapter on reverse genetics for avian coronavirus was published in 2017, and to follow those directions, you need very specific material. You could just focus on the buffers and mediums you need, such as OPTIMEM 1 with Glutamax-1, and do the work to see if indeed the lab there had those orders and you can do that with every needed material. There is a small possibility that the lab came up with some novel way of synthesizing genetic material, but that remains very remote. From there you can look for traces of plasmid vectors or other reverse genetics systems. Secondly, the furin cleavage site is an odd choice if the leak of the specimen was purposeful and intentional. Thirdly, in order to have a lab origin solely, you would need to amplify the genetic material through a process called serial passaging in order to recover high-titer stocks from samples. This process would likely delete the furin cleavage site, which would be evidence for laboratory handling. You would also see any evidence of animal adaptation in laboratory animal models, such as mice, but I don't believe we have that sort of evidence as of yet. We could also see evidence such as novel restriction sites, difference in DNA base content, differences in gene organization, selection markers, and foreign nonviral sequences. Lastly, from some recent research I read, I believe the first cases were clustered around the wet markets in Wuhan, which I believe are 9 miles from the Wuhan Institute.

There are a lot of arguments you can develop about why this isn't a man-made virus, and the zoonotic spillover event needs something like a wet market in order for the spillover to occur. That said, each time I look at the evidence, I vacillate between thinking it is a possible lab leak due to improper handling of samples, which could be possible given the relative newness of the lab, which would be an accidental situation that isn't necessarily a leak. That situation suggests that the virus existed in the population well before the pandemic began, and the process of serial passaging made it more robust relative to other Sars viruses. The other situation I think about looking at the evidence is indeed the wet market spillover, which is a danger any time you have consistent human and dead animal interaction. I also think that some of China's moves in the aftermath were to protect the profitability of the wet markets themselves, but I have to investigate that notion further.
