As I always say in these threads - there is no right answer and you can tell the age of the posters by their answers
There are so many amazing solos out there, but if I had to pick just one...
Under a Glass Moon - Dream Theater (John Petrucci)

Here we go . Best Solo!
All in with David Gilmour on several songs.
I am sure Zakk Wilde may have better ones but No More Tears has always rocked for me. Comparatively short to some others mentioned but yet intense.

RIP Ozzy.

Good choices. IDK what the "best" is and I don't have a single favorite. Some faves of mine not yet mentioned are:
Ancestral - Steven Wilson (Guthrie Govan)
Firth of Fifth - Genesis (Steve Hackett, written by Tony Banks)
Kid Charlemagne - Steely Dan (Larry Carlton)

This needs to be two categories: 1) EVH and SRV and 2) all others. Among the others, I agree with Allen Collins on Free Bird.
To color a little outside the lines, I'll roll with Shawn Lane on All Along the Watchtower. Some of you will have to change your votes on the other side.
There are two solos, one at about 1:10 and another at about 2:40.

SRV
There are two high notes SRV hits in Little Wing that are, to me, the closest thing to what a guitar is supposed to sound like in my mind.
My dad would listen to Walton and Johnson on his PC at the houhse and I more often than not would wake up to that solo.

Lots of really good answers here. I am going to drop a couple I don't see yet. Some of my own favorites are not necessarily technical marvels, but here goes anyway.
Man In The Box - the first full bit more technical solo I learned. I play it alot to this day. It's got alot of interesting techniques and that Cantrell flavor.
I Remember You - I've always loved Scotti Hill's playing, and this is one of his great solos IMO. It drips with emotion and ends with those harmonic bar dips that hands back off to Sebastian.
Sail on Soothsayer - Not to be confused with Soothsayer, which is a great song too. The whole song essentially is a bit of a softer melancholy solo.

Ritchie Blackmore's iconic solo on Highway Star (starts around the 4 minute mark)

Most of my rock faves have been mentioned. So, let’s delve into Funk, Funkadelic to be specific. Maggot Brain : A 11+ minute trip into the cosmos, piloted by Eddie Hazel! You can skip to the 1:15 mark if you don’t want to hear the intro

Going to switch to jazz for this one. The Ramsey Lewis Quartet doing a completely different take on Sun Goddess . No horns, Ramsey doesn’t do a solo, and Henry Johnson shows us mere mortals how it’s done on jazz guitar. His full solo begins at 1:58 and it’s Henry’s track from that point forward:

I much prefer the Cemetery Gates solo to the Floods solo.

Is it a solo if two guitarists play on it? If so I nominate Duane and Dickie on Blue Sky.
That's a great one. And 3 minutes long

This is such a subjective question. There are infinity shredding-style solos out there, and it's impossible to say which one is truly the best. So I'll go a different route and nominate this one. It is the closing song off an album centered around the concept of heroin addiction, and specifically how it is a closed loop - the addiction feeds back into itself, despite the protagonist's desire to be free from it. As the final song on the album, it represents the point at which the end becomes the beginning and everything starts over again, and the guitar solo truly captures that feel. There's no shredding here, just raw emotion. Probably my favorite solo ever.
