I have two that stand out.

At the end of Anger Management, Adam Sandler's anger management friends throw a surprise engagement party, which includes John Turturro jumping out of a tree and hammering Adam Sandler. 13 year old me lost it at that.

Funniest moment in any movie.... "Schmidt fricked the captain's daughter!"

The build up until Channing Tatum figures it out sells it.

There's like 5 on my list that have already been named, but the one I laughed at the hardest was the chest waxing scene in 40 Year Old Virgin. The whole movie was hilarious up until that point but that scene kind of came out of nowhere because his character was so quiet and lame then all of a sudden "ohhhh you FRICKER!" . I must have rewound that scene twenty times before I even finished the movie

Not really even a whole scene. For that, it's probably Springtime For Hitler from The Producers (the original, of course). And an honorable mention for LSD's audition scene.

This movie is full of hilarious scenes. But this has to be the best.

Another one from that film, when Stiller's character "cleared the pipes" before his date with Mary and he can't find the...ejaculant, and he opens the door for Mary and it's hanging from his ear and she uses it for hair gel. Not sure I have laughed that hard in a theater, ever. It doesn't hit as hard as some of the other jokes do on rewatch because the surprise isn't there, but man, on first view it was outstanding. I was on a first date, too.
