I keep a 38 in the night stand, 12g under the bed.
Wife can handle the 38 no problem, so in the event SHTF she would be holding it.

Anything that is reliable and you can most naturally hit your target with (which does not necessarily equate to "comfortable").
Ask people to let you fire their weapons in exchange for a box of ammo (if you can get any).
Are you set on a pistol platform? For an inexperienced shooter to use for home defense, my order of preference of platform would be:
1. Carbine
2. Pistol
3. Shotgun
But try each with two targets, and see how well/quickly you can methodically get two shots on each target is timed succession (without rushing).
Once you're set on the platform that works best for you, then worry about caliber.
Same selection process applies with caliber...whatever you can accurately send both first AND follow-up shots.

Strictly for home defense? 12GA

That would be a 9mm bullet and a 40 case, which equates to 357 Sig.

Not .40 that's for damn sure.

Sig P220
.45 ACP
(ETA: And I concur with those who might suggest you arm your home with a long gun, and my preference is a 12-gauge, pump action shotgun by Remington or Mossberg. A handgun is an inherent compromise. Portability is its only true strength, and that is negated in a defense of the home situation.)

Whatever you do, don't go out and buy a .44 magnum and take it to class thinking you're going to shoot 200-500 rounds out of it no problem.
Use a .22 or .32 for the class. Yeah, you'll get some laughs in the morning, but you'll be the one laughing in the afternoon.

They provide the gun from what I'm told... we pay for ammo and the class
This is thw best way to go about getting familiar with a handgun right?
Take a class?

#1 one shot fight stopper in all of handgun history.
Interesting ......... but (perhaps) a tiny bit misleading. Revolvers and semi-auto handguns (under .50 caliber) are unlikely to be reliable as "one shot fight stopper."
Plan on multiple shots -- and feel thankful if one shot takes care of the threat
