Either going to keep my current Sierra to have a truck available or go with a Denali. This question is geared at keeping the truck and having a separate commuter vehicle.

Let me know when you find a vehicle that checks all those boxes. Thats a tough one...
Resale is gonna take a huge hit due to miles.
Smooth/quiet is difficult to find with high MPG.
How much space do you need?
Obviously, an EV is out of the question.

Buick Encore GX. It fits the bill for all of your criteria. There are several different trim levels you can get to your liking. My kids each have one, and we have had very little issue with them.

Get a Subaru.
More reliable than Toyota and great ride and you're under the radar. Ain't nobody gonna steal it
I also like the CT? Cadillacs. Th 5 or 6 is very luxurious and smooth as butter.
The SUV/pickup baws in this thread are missing that part where you're going to be spending a lot of time on the road and you don't need the crappy gas mileage or bumpy ride.

Don't need to impress anyone? Toyota Crown for your criteria
Is your "space" criteria for something clean, like tech gear or medical equipment, or is it something dirty?
If the latter, maybe a Ford Maverick?
Really need more info. In my prior line of work in the auto industry, I needed a back seat and something that made an impression/conversation starter. The Pontiac G8 GXP was a perfect fit for that, and the next step on that same ladder was an AMG E-class or Caddy V series. The other extreme that got about the same results was a first gen PHEV, the Ford Fusion Energi, I would never had driven something so generic as a Crown to those types of meetings.

Get the Denali.
We just got one with the ultimate package and it’s an amazing vehicle
Incredibly smooth drive as well.
The hands free cruise control is badass

Space would mainly be for luggage, computer bag, and golf clubs. Occasionally take a small group to lunch or dinner. Doesn't have to be cavernous, but comfortable for back seat riders would be nice.

I have had 3 Lexus ES as business travel cars. They all saw extensive miles with almost no repairs. The current one has over 460k miles, and the total of all repairs is under $2600, none of the failures stranded me.
It is the perfect solution to your "problem".

Yeah, I know all 5 is tough, so ride and reliability are first and foremost. The rest are really gravy.

Camry, Corolla, Accord, or Civic. I'm assuming you're getting a vehicle allowance?
ETA: since you said ride is most important, I'd say a Camry or Accord.

I’ll say, I rented a Honda Civic this week. It was pretty zippy so prob souped up a bit but very nice. I’d buy one tomorrow if in the market. Best rental I’ve had.

You want as little maintenance as possible and something lightly used so you don’t eat depreciation. Also, I like 6 cyl for durability and leather because if you’re going to keep it until the wheels fall off you’ll wear out fabric seats with a lot of windshield time.
Carollas, Accords, Civics etc. are great but they get small on long rides and can’t handle back seat passengers well.
An Avalon fits but there aren’t a lot of them out there. By the time you find an Avalon or Camry with 6cyl and leather you’ll spend about the same as you will on an ES350.
Just get ready for what you want to do with all of the extra money that you’re used to spending on maintenance.
ETA: after typing this I’ll probably blow a cylinder on the way home.

OK, so 4-door something for sure...
Kinda leaning to Accord/Camry.
Bulletproof cars, rides well, quiet, excellent resale even w/ high miles.
