Most I've ever laughed at a movie in a theater, so much so I was hurting, was the original Jackass feature film.
Borat is in that same vein, it was as shocking as it was flat out uncomfortable and laugh aloud and outrageously funny.
For traditional films, I don't think Blazing Saddles can't be topped.
But for single sequences, I'll put the Pink panther "paralell bars" as pretty much the height of movie comedy. LINK

Cop Out had me laughing hard in the theatre. Haven’t rewatched it since but can remember hurting from laughing.

Very old school, but “It’s a Mad, Mad, Mad, Mad World”. Everybody is in it, and it still holds up reasonably well.

I hated Hangover. But I wasn't born in the 90's either.
To this day, I still laugh every time I watch Coming to America. I sometimes laugh just thinking about some scenes.

The hardest and most continuous move inspired laughter I’ve ever had was as a 14yr old watching Happy Gilmore.
