Changes over time.

As a kid, it was easily Airplane. And then in H.S. Holy Grail and British comedy became the thing.

Then the 90's classics.

Later, Dr. Strangelove.

It's dumb and dumber for me.

Additionally, this thread makes me sad about the current state of comedy - DEAD.

Every summer I used to look forward to the new comedy blockbusters coming out in theaters.

Not anymore.

I'm afraid that I'll never again laugh until my face hurts in a movie theater.

Pure Luck with Martin Short, because of this sequence.

My younger brother and I were watching with my mom and dad when it first came out on VHS. He gets stung by the bee and starts to swell. My mom got to laughing so hard she choked herself out because she couldn’t breathe. When she recovered and came back around she had peed her pants also.

The Big Lebowski. It’s still funny to this day to me and it always will be as long as God makes me stay on earth. Fifty years from now it will still make me laugh.

I could probably list 50 great comedies, but Airplane, The Naked Gun, and Top Secret still make me laugh consistently with repeat viewings...

Some that haven't been mentioned that also cracked me up were 40 Year Old Virgin, The Muppet Movie and Napoleon Dynamite. Seems like most people on here hate ND but it's one of my favorites and its extremely quotable.

Loved Blazing Saddles, The Jerk, and Airplane from back in the day. Lots of other good ones already said.

One I didn’t see mentioned that had me rolling in the theaters was Tommy Boy

Hot Rod is one of the most underrated funny movies ever. That movie makes me crack up every time I watch it

Christmas vacation, both caddyshacks, step brothers, talladega nights, the jerk (I FIGURED OUT WHAT SPECIAL PURPOSE IS FOR!), dumb and dumber...all are up there. The wedding singer is a good one too. ETA The Other Guys. Laughed almost all the way through it.

I’ve always felt the original Ace Ventura was somewhat overrated. When Nature Calls, however, is probably my pick for the answer to the OP. The rhino scene alone would have it in the top 5. I’m laughing right now just thinking about it.
