If we are going by the book, then it is Mufasa easily, because Shere Khan has a lame foot.
If we are going by the movie then I'd still give the edge to Mufasa just because we know Mufasa is battle tested.

If we are going realistic then Shere Khan would rip Mufasa apart in fairly short order

Some years ago I read about an effort to re-introduce lions into some area where they had long been extinct, maybe in India, can't remember. Tigers had never been known to hunt in packs but apparently they will when you introduce lions into their habitats. The project went on for a couple of years but the tigers kept killing the lions within days of release.

Male lions don’t hunt with the pride. If they don’t have a pride, they’re hunting alone (or with another male lion) or scavenging.
