Dumb & dumber. I was 5 or 6 and it had fart jokes so yeah it had me.
For more recent, though not in theaters and very random, Scary Movie 3. When Anthony Anderson pumped the shovel like a shotgun with the shell coming out I lost it.

ok i just remembered mine...Harold and Kumar escape from Guantanamo Bay when the agent pours the grape soda out in front of the black dude to try to get him to speak. There were several black people in the theatre so everyone was trying not to laugh to hard but they had one guy just letting it all out which made me laugh even harder

Wasn't really a line, but I was high as shite and the hardest I've laughed was in the Avengers when the Hulk just picked up the bad guy and slammed him on the ground 10 or so times.
