I don't usually do "bests," but as to my favorite: Airplane easily.

Unless Princess Bride is considered a comedy in which case: Princess Bride, followed by Airplane

Next is Young Frankenstein

Of course if we were to expand to TV, my all time favorite comedy is Fawlty Towers. That show always had me laughing out loud.

Lebowski is my favorite, but not the funniest.

Holy Grail made me laugh more than any other movie on first watch.

Hot Fuzz and Old School are probably tops in terms of most laughs upon repeat viewings.

I'm partial to Major League....

but Jamie Lee Curtis topless in multiple scenes of Trading Places is tough to beat, if not impossible.
