We arrive on a Saturday morning after driving from the cruise port. Staying near the Pantheon. Saturday we have Pantheon at noon and Colosseum underground booked at 2:45.
Sunday we have a tour by train to Pompeii (daughter and mother in law really wanted to see it). That will take most of the day. Monday we have tickets to see Sistine Chapel first thing. What else would you try to reserve or make sure to see the rest of that day or Saturday after the Colosseum? We leave on Tuesday.

When we were in Rome, we did the following:

Toured St. Peters Basilica, Vatican Museum, Sistine Chapel, day trip to Pompeii, mass at St. John Lateran, Piazza Navona, Pantheon, Spanish Steps, Trevi Fountain, toured the Coliseum and attended mass in St. Peters with Pope Francis.

Awesome trip...

I greatly enjoyed a hop on/off double decker tour. Great way to see a ton of the city. That's beside the obvious forum/ colisuem and Vatican which you should definitely do, and which I'd advise buying tickets for ahead of time.

My wife and I just left Rome, and are currently in Capri.

In Rome, we found the best single thing we did was a private golf cart tour of the city. It was just us and the driver. He asked what we had seen and what we were interested in, then gave us a custom trip around the city. He stopped at a few spots for us to explore at our pace, or just drove past some spots that we had already seen. The total trip was 3 hours, and that was perfect. It gave us a great starting point for anything else we wanted to tour.

It was “Turtle Tours”, but we booked it through The Tour Guy.

Pretty good list already. I'd add in grabbing lunch and walking around Trastevere one of those days. Your daughter and MIL might like window shopping on Via del Corso too.

Be sure to pop your head into every church you pass. The most amazing art and sculptures are behind those green doors.
Some of the best ones look like they are abandoned but are not.

This is great advice even for Protestants :) as it takes little time and there is priceless art everywhere. Some to especially be on the lookout for San Pietro in Vincoli, Santa Maria Trastevere, San Luigi De Francesi, Santa Maria Della Vittoria.
