Hard to choose because all have such beautiful scenery, but I think #1 would have to be the Overseas Highway to Key West.

Others that are a very, very close second...
US 34 up Big Thompson Canyon to Estes Park
Utah Highways 12 & 63(?) through Red Canyon and into Bryce
I-70 in Utah between Green River and Salina, across the San Rafael Swell.

I drove the Amalfi Coast road SS163 this past May and survived. Stunningly beautiful to drive.

Most fun road I've driven in was the Tail of the dragon at Deals Gap, NC.

Million Dollar Highway in SW CO, US-89 in Utah, US 395 in California, Tioga Pass Rd, and the PCH from Brookings Oregon down to Cambria CA all come to mind right away. US-50 through Nevada is really cool too, nothing but mountains and desert with the occasional village that has a gas pump every 100 miles.

ETA: the Cabot Trail on Cape Breton is out of this world too but gas prices will make your head spin

2. MDH from Durango to Montrose
3. Trail Ridge Road in Rocky Mountain NP
4. Can't remember the name Utah state road that goes from I-70 to Moab (not US 191 the other road east of 191)

Wife had to hit the travel booze bottles in the glove box

No guard rails adds to the fear factor as does the 50-60's era autos still laying at the bottom.

My wife was too freaked out, we only got to Silverton and she made me turn around back to Durango.

I’m going back one day w/o her.

The drive from Putgatory to Silverton was beautiful enough I cant imagine it gets that much better?

Was lucky enough to hit it at the height of the fall foliage though.

Nothing compared to the road going up Pikes Peak before they paved all of it.

In no particular order:

1) Sea to Sky (Vancouver to Whistler) although I am bias as I was born and raised there

2) Oak Canyon from Sedona to flagstaff (my happy place is Sedona)

3) Banff to Jasper (Highway 93 and such) - Stunning beauty
