Could he do that by being respectful and saying while there are many things he agrees with Desantis on and he’s a good young man, he disagrees with him on specific policy issues such as ____ and Ron just does not have the experience he does to get it done ?

Trump has a primary to win and it should come as no shock to anyone that he'd go after Ron! with everything he's got.
The funniest thing about this entire ordeal is how Ron! supporters are shocked Trump would be throwing the kitchen sink at him. He fights as dirty as humanly possible and had indicated he would from the jump. Ron now has to be destroyed and Trump will overkill just to completely neutralize him likely for the rest of his political career... But you knew it was coming, didn't you? Honest question: How could anyone not?
