As long as it isn't strictly limited to rock music, I think you could make a good case for Outkast, or even better Wu-Tang Clan.

I like Wu-tang, but I think you'd have a tougher time making a case for them. People that are really into hip-hop like Wu-tang. Outkast fans are a much more diverse lot. Basically, if you appreciate good music -- and you aren't one of the sad, backwards individuals ::cough:: rocket ::cough:: still mouting a futile crusade against hip hop -- you like Outkast.
Until somebody tops them, Outkast is my favorite group of all time, regardless of genre. I can go deeper with an Outkast playlist than I can with any other band/artist in the world. And I like to use that as my test. Also, ATLiens is my favorite album.

Yes, #1. Honestly, I don’t see who else has a case. (And let me first say that REM isn’t my favorite band, I’m not this dyed-in-the-wool REM fan who has an altar built for Peter Buck). If you took an objective measurement of longevity, commercial success, artistic influence, and critical acclaim – I don’t think there’s another band that scores as highly as REM on all four metrics.
Longevity – this eliminates CCR, Guns n Roses, the Pixies, Nirvana, and every other short-lived yet great band. I’d have each of them on the list, but their small catalogs would keep them out of a reasonable “best band ever”.
Commercial success – I honestly don’t care much about this metric, but I think the #1 band should at least have some chart success. This would eliminate bands like Fugazi, the Stooges, Pavement, and even Parliament. REM was one of the biggest bands on the planet for about five years, selling out stadiums, selling millions of copies, and enjoying one of the richest contracts ever signed. REM is one of the most commercially successful bands of all time.
Artistic influence – REM’s influence cannot be understated. They invented indie rock, and they are the dividing line between punk and post-punk. Who else invented a genre? Public Enemy pretty much invented modern hip hop. Metallica invented thrash metal. Minor Threat invented hardcore. OK, that’s simplistic as there’s always a 100 other bands, but REM is generally credited as being the fathers of “alternative rock”, which is a pretty big section of iTunes.
Critical rep – Nearly unimpeachable. Even the Beach Boys have that 20 year period without Brian Wilson we don’t like to talk about. Metallica squandered their goodwill on the Napster wars. The Allman Brothers are beloved among their own corner of the world, but generally ignored by “post-rock” or any alt-rockers. They are traditionalists, which is cool, but they are a throwback before 1977 and stand as a huge boulder in the development of pop music, fighting the stream (this isn’t a knock – I love the Allmans). Steven Hyden invented the Test of Five. You can tell a truly great artist by their best run of five consecutive albums. Very few bands released five consecutive great albums: Murmur, Reckoning, Fables of the Reconstruction, Life’s Rich Pageant, and Document are all touchstone classics (except maybe Pageant, though it is one of their most popular albums).
Really, I think REM’s only competition for best American band using an objective rubric is either the Beasties or Sly and the Family Stone.
