These are no real actors but portrayed the actors and actresses in the movies.
I'll open with "Kirk Lazarus" (Tropic Thunder)..5 Time Academy Award winner!

I was surprised RDJ actually got a Best Supporting Actor nomination, not because he didn't deserve it- he absolutely deserved it - but they never give comedy awards. Of course, Heath Ledger won it that year (posthumously) for the Joker. I can't really disagree with that decision either.
Ones off the top of my head:
Nic Cage in Unbearable Weight of Massive Talent.
George Clooney did a great job as Jay Kelly
Man on the Moon - Jim Carrey as Kaufman
RDJ as Chaplin

Dame Maggie Smith played a character named Diana Barrie in Neil Simon's California Suite (1978). Sharing many of her scenes with Michael Caine, she won the Oscar for an Oscar-nominated actress (certainly the first such performance).
Also, somewhat ahead of its time, Caine plays a not-so-closeted antiques dealer Sidney Cochran, whom his wife, Maggie's character Barrie, functioned as a beard.
Most of their scenes were light argument/banter that worked particularly well in the anthology style film.
ETA: IMHO, this is the best performance by an actor playing an actor, with all due respect to RDJ's Kirk Lazarus and Leo's Rick Dalton.
