It's not, and thusly says so in the OP. It can be longevity or iconic stature or whatever.

Lucille Ball is a lock from the early days of TV
Archie Bunker from All in the Family. Was the most popular show in the country for years and also had more political content than a lot of earlier shows.
Heathcliff Huxtable (Bill Cosby) was very popular for years and one of the more important AA roles on TV
Homer Simpson I think is more representative of the Simpsons than Bart (the show started more about Bart, but became more about Homer).
Seinfeld was arguably the greatest sitcom ever. George and Kramer are wackier, but the show revolved around Jerry.
Tony Soprano is arguably the icon for the modern golden age of high production value cable productions
Honorable mention to Sheriff Andy Taylor (played by Andy Griffith) was incredibly popular in his day

I'm not exactly a connoisseur of pop culture and all-time tv shows, but when I first read this thread title these 2 names came to mind:
Homer Simpson
Jerry Seinfeld
Also, Lucille Ball and Tony Soprano are solid picks for the other 2 IMO

was my first thought.
I don't know if it can be narrowed down to just 4 characters.
There's just too damn many of them. How can you leave off the Fonz? or Andy Griffith, or Archie Bunker, or Granny Clampett, or Hawkeye, or Mr. T, or Marshall Matt Dillon, or Boss Hogg, or Optimus Prime, or Dave Chapelle's Tyrone Biggums, Clayton Bigsby, or slutty Carrie, or even Dr. Sheldon Cooper?
