I'll go off the reservation and say Chase. In a movie where all the other comedic characters were over the top in some way or another, he played it completely deadpan. His understated delivery really played up the sheer ridiculousness of his lines.

Only scene that was anywhere near his usual over-the-top slapstick was his date with Lacey. And it actually worked there.
I'd be hard-pressed to name a single funniest character. The whole cast made it work.
There's a 2009 doc about the making of the movie, Caddyshack: The Inside Story, which is worth checking out.
Unfortunately Sarah Holcomb remains an enigma.

Supposedly all the drugs on the sets of Animal House and Caddyshack were her downfall. She was only 18 or so for Animal House.
*For the longest time I did not realize that was the same girl in the two movies. That accent was a boner killer.

Dangerfield followed by Knight for sure. But I really also love Chevy Chase.

Yeah he dropped out of high school early to enlist for World War II and served in the European theater. I’m sure there were several moments where he showed up to shoot and one person opposite to him was hung over and the other was still asleep in their trailer and he thought “I faced bullets to save this?” Lol

I think it’s Ted Knight. Dangerfield was hilarious, but he’s a one trick pony.
Knight actually gives a great performance and his quotes are just about as good.

Has to be Bill Murray for me. Every time I've ever set foot on a golf course the Spackler quotes come out. Really, any time you have to swing a club-like sporting instrument. Baseball bat, croquet mallet, polo.

Knight is the central antagonist for each of the other comedic or straight characters: Chase, Murray, Dangerfield, and Danny. And he's amazing because he's entertaining and substantial while playing the foil for all of them.
