Tomei really stood out in a great movie. That is damn impressive. There were a ton of laughs in My Cousin Vinny. Just a tremendous job by Tomei in that movie.
Kahn was great with a director who knew how to get the best out of her talents. Kahn + Mel Brooks = comedic gold. Her scene from History of the World Part 1 where she's picking out her escorts to the orgy is drop dead funny.

Heather Grahams cameo in Anger Management is an absolutely hilarious bit. Prolly doesn’t count, but it’s great.

Agreed Marisa Tomei in My Cousin Vinny is #1. McCarthy in Bridesmaids is probably #2. Rose Byrne also kills in Bridesmaids. Charlize Theron is great in Young Adult.
However, the only female comedic performance approaching Tomei would be Julia Louis-Dreyfus in Veep. Obviously different platform. But the only thing as iconic.

Kathleen Freeman - Enid Borden in Dragnet
This scene makes me lose it every time.
I don't even care that Freeman's nowhere else in the movie. She creates a character as instantly memorable as anyone else in the movie, in less than two minutes of screentime.
