Here’s one from way back: Jack Lemmon as Professor Fate in The Great Race.

What a great vintage comedic villain ! I love that movie. Peter Falk as his sidekick/henchman made this one of the greatest slapstick melodramas ever.
Here's another oldie:
Dr. Goldfoot and his bikini machine

ben stiller was basically the same character in heavyweights and dodgeball. and it's definitely one of the first that comes to mind. there are some differences, but they're very similar
There are exactly the same character in my head cannon universe. Obviously he had to change his name to White Goodman.

All great ones listed thusfar.
I'll add these...not sure if they were going for comedy, but it sure came out that way.
And for your troubles, Julie Newmar.

Julie Newmar was my crush back in the day. I still love brown-and-brown.

Are you still holding on to all the anger? I said I was sorry, I meant it.
Also have to put Adam Scott as Derek from Step Brothers up there as a great comedic villain. DANE COOK PAY PER VIEW

“Why don't you go eat that outside and then come on back in.”

Even though she's got to be in the late 80's I probably still would just to say I nailed Julie Newmar
She did a sexy picture celebrating her age recently:
