Definitely go 9mm.... Hollow points for home defense, cheap stuff at the range.
I'd take the Walther ppq if going polymer. Much better trigger than a glock, so it's more fun to use on the range.
Personal preference would be a steel gun, either cz75 or sp-01. Sp-01 may be a tad heavy for your wife though. I had a p01 and wasn't a huge fan, the smaller size and alloy frame wasn't particularly appealing.

I tried to post this fur you last night but I found a gun for you on palmetto state armory. Their service is horrible but cheap as dirt. They have a Smith and Wesson 9 mm 1911 like gun for 269. Four inch barrel which I am not big fan of but u can't go wrong at that price

I absolutely love, and i mean love my ruger sp 101 357 magnum. Keep it loaded with critical defense. Shoots 38's great also.
