4.9 Magnitude Earthquake near Iran's Nuclear facility

The short answer is yes: a nuclear explosion can cause small earthquakes. But it is unlikely to affect the earth's tectonic plates or cause a volcanic eruption. Although a nuclear explosion releases a lot of energy in the immediate region, the amount of energy is small compared to other stresses on tectonic plates.

There was a 5.1 there on 12/27.

Active area?

25 earthquakes in the last 6 months of a 4.0 or higher.

They also have a unique signature unlike natural earthquakes.

It appears so. Looks like the middle of Iran is fairly quiet, but all of the borders are seismically active.
