The two peoples are ethnically and linguistically "siblings" - they are Western Slavs and the current ethnic fault line comes from the time where they were part of the "dual monarchy" Empire of Austria-Hungary (with the Czechs falling on the Austrian side of the line and the Slovaks on the Hungarian). The languages share a high degree of mutual intelligibility with each other (and Polish).

The Czechs are somewhat more "Western/Central" (meaning German influenced), while the Slovaks are more "Eastern/Southern" (meaning Hungarian influenced). The Czechs, largely due to German influence are more Protestant (and largely irreligious in the post-Cold War era), while the Hungarians are largely Catholic.

The Czechs are more urbanized and the Slovaks are more rural.

There are other differences, but that's a good start.
