Several in Zoolander:

John Voight as his father
David Bowie as the walk-off judge
David Duchovney as JP Prewitt
Justin Theroux as the Evil DJ
And, of course, Billy Zane

His intro in that movie is art.

Joe Pesci, appearing in a Bronx tale

On the initial viewing, you don’t think much of it, but at the end when you realize who it was and that it definitely wasn’t over a parking space. I thought that was impactful for C’s story.

are we talking impact on the plot? or just a generic impact, like it's a popular or funny cameo?

i think this is a good answer. i am seeing a lot of answer that have misinterpreted the question. Confusing most impactful with most memorable.

this character impacts the entirety of the rest of the movie despite being in it for a few minutes.

Baldwin in Glengarry and brando in apocalypse also good answers.

i would submit keaton's Beetlejuice. He has like 15 minutes of screen time the whole movie but steals every scene and is the whole driving force of the narrative.

Chuck Norris in Dodgeball. They don't get to play for the championship without his vote.

Lance Armstrong in Dodgeball. They don't get to have Chuck Norris vote for them to play the championship without him convincing Vince Vaughn to return to the team.

Rip Torn in Dodgeball. They don't even know how to play dodgeball without his brief stint as coach.

Dodgeball was a great movie.

Beiderman in the movie Deep impact.

when the comet split in 2, you didn't see much of Beiderman - but it sure had an impact
