Way too many wonderful scenes to come up with a best ever, but the first one that popped into my mind:

Breakfast at Tiffany's: Paul giving Holly his cashier's check for $50 to go to the powder room in the library.

YouTube Link

One of those moments where you have to shed a man tear and give a slow clap.

Lots of great scenes mentioned in this thread already, I particularly agree with the Godfather, Dead Poets Society, and Gladiator. A few more that I don't think have been mentioned:

Classics
To Kill a Mockingbird: Scout, you stand when your father passes.
Casablanca: Playing the national anthem

Action Scenes
The into to Raiders of the Lost Ark
Staircase scene in the Untouchables

Father & Son scene at the end of Field of Dreams.

This is probably mine. I've always wondered if John recognized Ray at the end. It seemed like he choked up a bit after John called him Dad, but it wasn't clear cut in my eyes.

The scene that showed how Andy escaped in Shawshank Redemption is my favorite by a huge margin.

It's an odd (and emotional) scene for sure...John certainly doesn't seem to recognize Ray at first.

At first John asks Ray if he's in Heaven, to which Ray responds, "This is Iowa." John says, "Could've sworn this was Heaven."

After that, Ray ask John if there is a Heaven, to which John replies with absolute certainly, "Oh yeah! It's where dreams come true." Therefore, in that moment John tells Ray that Heaven certainly exists even though a moment before he asks Ray if he's in Heaven.

Ray takes in the moment, glances at his farmhouse where his wife and daughter are rocking in chairs, realizes that everything is perfect and then he says, "Maybe this is Heaven."

Soon after this scene, Ray asks John, "Dad, wanna have a catch?"

And John replies (with tears in his eyes), "I'd like that."

So, it seems like John knows exactly who he is playing catch with at the end of the scene even though at the beginning of the scene he acted like he didn't.

Odd scene. The oddness is what makes it ghostly. It's what makes it masterful.

It's the perfect scene.
