The most nightmarish issue I fought with was during my first build. Wouldn't post. I probably rebuilt the thing 3 times and reseated components and connections over and over before finally deciding to go through the mobo manual line by line.
The mistake I'd made isn't even a relevant mistake anymore. There used to be a jumper that told the MB to run the Pentium 4 at either 100 or 133MHz FSB (with a fixed multiplier). Jumper was set to 133 by default, but my CPU was based on 100.

Problem was I had already mounted everything and the cpu cooler was blocking it from view.

I've used Macrium Reflect usually, but MiniTool partition wizard now has a built-in clone feature that works well, too.
EDIT: Those aren't open source, but they are free. Open source... there's clonezilla. I've never used it before.

So I am plugging in my USB 3.0 flash drive into the USB 3.0 port on my mobo and still getting 16MB/S transfer speeds.
There are no USB drivers on the ASUS website for my board.
What am I missing?

Bottleneck is the flash drive itself. Just because it's USB 3.0 doesn't mean it will even be faster than USB 2.0. Read the reviews on that flash drive. People citing 13 MB/s sequential write speeds on average.

Sandisk Extreme flash drives are probably still on top. Maybe some others out there, but the Sandisk/Toshiba factory is probably supplying the chips. You might also look into compact SSDs if the price is reasonable. I have a random collection of flash drives, some USB 3.0, but I didn't buy any of them with speed in mind. I have a Sandisk ultra fit that does about 30mb/s sequential write, but mainly I like that it's 64gb and about the size of a multivitamin.

I did lose it for about a day. Fell out of my pocket and under the seat in my car. They're nice if you intend to keep them attached to a laptop. The one you linked is what I had in mind for speed.
