Chicago - The Great American City

Boston - The closest thing to Jolly Ole England you'll get

Las Vegas - Adult's Playground. It has it all.

Key West - When you want to get away and enjoy the tropics

Los Angeles - "La La land", Hollywood, Malibu, Venice and Muscle Beach

Some other cities worth checking out (since I think most people would have a consensus of a top 10) for a long weekend.

1. Louisville (great DT; unique city)
2. Kansas City (good DT; BBQ)
3. Cleveland (Rock and Roll HF is amazing)
4. Savannah (great DT; history; food)
5. Baltimore (good DT; history; day trip to DC)

I am surprised that Orlando and Austin are not mentioned more.

Love me some Venice.

My new favorite watering hole, Venice Ale House, is right on the beach and I really enjoy walking the Canals Walkways.

Always have to stop in front of Hank Moody's house on Brooks.

I was going to mention Kailua, but it's definitely not a "big city". Same with Savannah, I'm not certain that it would be classified as "big".

My list:

NYC
Kailua
DC
Savannah
Denver (and surrounding areas)

I'm pretty sure that says Venice, LA...not Venice, CA

Venice is a neighborhood in Los Angeles, so figured that's what the "LA" abbreviation stood for.

Somebody downvoted because I said Houston

I didn't down vote but if I had it would have been because of Indianapolis. I have a hard time understanding how it would make a top 5 list.
