tell her someone is moving in on that day and she better be out or sleep on the floor in the hallway with the rest of her stuff.

No the OP said she was renting a room, not renting a room in her own house.
It makes a difference why she needs the room renter to leave, because if its for a good reason like the house is being sold then she can push her luck and not do it 100% legally. Even though she only rents a room she still has tenant legal protections.
So if she just wants this room renter gone for a dumb reason like so a new friend can move in, she should go through the proper legal method and file for an eviction and give the legal notice. It would be stupid to risk legal ramifications for a dumb reason to leave.

It's to sell the property. It's half occupied by my sister half by the two tenants.

Is this in Louisiana?
A LA lease can be unwritten. Unless otherwise agreed, it is presumed to be month-to-month.
The tenant or the landlord may terminate the the lease, for any reason, with ten days written notice before the end of the month. The 5-day notice is used for non-payment of rent.
This info, plus how to deliver notice, how to count those days, and the eviction procedures (if it comes to that) can be found in a pamphlet at the Attorney General's website. LINK
The more notice you give, the more likely they have time to get their shite out without having to go to court.
