Male side:
The Duke - 50 years, ~ 170 films - A lister for over 40 years of that
Eastwood - although he transitioned and has been behind the camera for much of the last quarter century - we're still talking about a guy who had lead roles from 1964 to 2012 - and counting.
Stallone/Pacino/Deniro/Brando - kind of make up the next tier - and all Italians - perhaps a 5th to include here would be Dustin Hoffman?
Honorable mentions:
Olivier - overlooked because of his stage work and British Cinema - pretty much an A-lister for almost 60 years.
Welles - was a major force while still in his 20s. His late career swoon probably keeps him out of the running.
Females:
Bette Davis - 53 years as a screen actress, 10 Oscar nominations
Stanwyck (the heavy television at the back end probably dims her star a little) was on screen, big and little for a long, long time.
Meryl Streep - probably still behind Davis, but has the Oscar credibility (17 noms, 3 wins) and still a viable actress in her 60s. Julia was in 1977, so we're at 36 years and counting.
Lucille Ball - purely from a television standpoint, Lucy was television for almost 30 years, and remains synonomous with sitcoms and syndication. Probably the most influential star on this list, save perhaps the Duke and Clint, and is the single most important person in television history.

For a woman Streep has no peers. She has been great for a tremendous length of time in a profession obsessed with youth.

Meryl Streep, Jodie Foster, Drew Barrymore, Kate Winslett, Christian Bale, Shirley Temple (as an ambassador), Marlon Brando, Robert Redford, Christopher Walken, Michael Kane,
Those are all I can name off the top of my head. They were able to act at a very young age and continue their success.
