In terms of all peaks, no valleys pure Jokes/min:

Tropic Thunder
Walk Hard: The Dewey Cox Story
Talladega Nights
Wedding Crashers
Wet Hot American Summer

are in a tier by themselves.

There are some strong contenders for comedies that are overall better, more cohesive films, but none that I would deem "funnier".

Watching Hangover for the first time without any real heads up or spoiler is up there.

I’m not sure that experience will be replicated. There may be funnier movies, but not like that.

I think that Walk Hard was funnier the first watch through over the Hangover, but once you know the direction the Hangover is going I do think it becomes less funny. Caesar’s Palace would have certainly figured out what was going on at worst within 5 minutes of Doug throwing the mattress from the roof, and if you’ve ever lived in Asia, you’d immediately realize how dumb the ice machine plot device was in the sequel.

But I appreciate comedies more that get funnier once you know the direction they are going with. The first time watching Hot Fuzz, the first 2/3rds of it isn’t as funny since you’re intentionally being misled all while subtly planting the seeds for the last third of the movie. Some of the punchlines are more set up in the last 1/3 part of the movie and delivered in the first 2/3rds, something you will only get after seeing the film multiple times. That is a type of comedy that clearly trusts in the intelligence of its audience and that it’s good enough to be viewed multiple times. There aren’t jokes like this in The Hangover, while that’s most of them in Hot Fuzz and The World’s End.

Well, at least the film industry has 80 more years to come close to movies produced in the 1900's, and by their production so far, they'll need them.

Bad Santa gets my vote for the funniest.

Anything else worth mentioning has been mentioned. It's been a pretty mediocre century for Comedy and the past decade was particularly empty. Given the current political climate, Comedy seems to be dying, which is a shame.
