I know he's not a true comedian, but a lot of his roles were the goofy character in Adam Sandler movies.

I remember watching it when it came out (as a kid). I wasn't really that moved by it. Need to watch it again as an adult to see how it resonates now.

I'll mention a couple that I haven't seen brought up yet:

Richard Pryor in Blue Collar
Albert Brooks in Drive
Jonah Hill in Moneyball
Patton Oswalt in Big Fan

I think part of the appeal a comedic actor brings, is that he can already make us laugh. That plays to emotions, something important in connecting to characters.

Think about real life people you know- the ones that have a smile on their faces, that can make you laugh, you generally like these people more, and know them more personally, than the stiff folks.

So when you see a comedic actor in a serious film, you usually already have a liking to him; and if he's never in a situation to be relaxed and silly, you have a little more empathy to him... subconsciously you might think "man, I wish things would work out so he could lighten up".

I forget the movie (it was just 'ok'), but Steve Martin did a romance flick once. He was flirting with a gloves sales chick in a Macy's or something like that.

He played a single, sort of lonely older guy, and I just remember that he was excellent in it. Extremely expressive, but very understated performance. Really no comedy in it at all.

I think you're talking about "Shopgirl," with Claire Danes. He apparently wrote it too, which gives it a somewhat self-indulgent aura in retrospect. Yeah, it was just 'ok' but I'll give Martin credit for pulling off the serious. He's no Bill Murray.
