These are the top 3. If you ask any comedian who is/was their biggest influence it's Pryor.

I saw Carlin live. I'd put him at the top. I saw Eddie Murphy live as well, he's up there.
Seinfeld and Leno, at their peak, were up near the top. One of the worst things to happen to stand-up was Leno re-tooling himself to become non-threatening to the public. I saw Leno in Baton Rouge at the old Funny Bone in '86 when he was THE guy to stay up for on Letterman. He was hilarious.
Someone who should be mentioned frequently in this thread is Steve Martin. People forget that prior to his SNL and movie careers, Steve Martin was the biggest stand-up in the nation.

I would probably go with him but also
Richard Pryor
George Carlin was pretty fricking great
As was Mitch Hedberg. I could make a case for any of these
Oh also, some of Jim Carrey's early stuff is absolutely insane, in a good way

I think it's because he is exceptionally intelligent. There aren't many dumb people that are really funny. Takes a creative mind to put things in a way that others haven't thought about, or have and didn't consider funny before. I fricking love comedy

Honestly, I think it's George Carlin because of the way he created the way comedians work to this day. Do the special and then start with a blank page the next day. Spend all year building a set, do a special, and then start over again. Before Carlin, comedians tended to stick with routines for years, even decades. TV changed all that, and Carlin was the guy who truly changed the game. He also was a master of language.
I tend to like the "outlaw" comics: Bruce, Hicks, Stanhope, Bamford, etc. but I think i'd have a hard time calling them the greatest. They are important, but there's something to be said for being able to break out of the niche of comedy nerds.
Right now, I'd say Louis CK, Chappelle, Oswalt, and Rock are the tops of the game. I'm with y'all who miss Giraldo and of course Hedberg.
Also, points to Kathleen Madigan for just being a great technician. She's just flat funny.

Some of Jim Carrey's finest work. 5 mins
do yourself the favor and watch this. So classic
"cuz that's what my life's like.... CRAZY THINGS HAPPEN TO ME"

Stand up I think is one of those things that it's damn near impossible to name a GOAT. There are OG's, but there are so many that people connect with in different ways, it's very subjective

I refuse to claim a "best," but there is a definite top tier which has to include Hicks, Pryor, and Carlin. Eddie Murphy was damn good back in the day, too.

Yeah, and different people are trying to do different things. Tig Nataro's Live set is already legendary, but it also doesn't have any jokes. It's just a person who is almost literally at the end of her tether. I wouldn't recommend it to just anyone (but if you're a fan of standup, go find it). Meanwhile, stadium comics don't do much for me, but clearly appeal to a lot of people.
Which is why I went with Carlin, because he wasn't just funny, he changed the way comedians work.

All time: George Carlin. Man do I wish he was alive to see the election last year.
Currently working: Bill Burr. I think he's passed Chapelle.
Bill Burr vs stupid feminist the night of the election: LINK

Robin Williams has to be up there as well.
My wife didn't like the new Chappelle special in L.A., but I thought it was great. He's right back close to his old form if you ask me.

I didn't find him particularly funny when I was younger and he was still alive.
But as I got older his material definitely grew on me.

ETA: To be fair I thought the Chappelle show was hilarious. I just don't find his stand up to be that funny.
