this one is big ... but for a regular season game there has been none bigger than Florida 2007.
Now IF we win Saturday and run the table then play for the BCSC == in reflection this one might be the biggest regular season win of the era.

Your problem is, you can't see past your own nose, and that's fine.
Is the BCSCG a big win? Yes. It's always important to win championships and of course the BCS is the grand daddy of them all.
But I'm talking about in terms of magnitude of the games itself relative to the level of competition and what it means to the program.
A) The BCS game, whether rightly or wrongly, was considered by most everyone a game that we should have won and blown out. Secondly, most believed that Miles won this game on the merits of Saban's accrued talent, not through any coaching merit of his own. It was also a home game, more or less. In that sense, the "bigness" of the win is diminished.
B) If we were to travel to a tough environment, beat a team that is thought to be not only better, but demonstrably better, than us, and with Miles own players/coaches etc. it will be perceived as a much bigger win.
It would also go a long, long ways to showing that LSU is not slipping down the ranks of the SEC, which is very important right now with the dominance of Florida and rise of Alabama.
That is why I say this would be the biggest win.

You have to separate post season and regular season when assessing something like this, because any championship game (SEC or BCS) is more important than any regular season game.
You also have to separate if you are going to gauge the importance of the game in hindsight when the whole season has been played, or exclusively at the time of the game. Of course the later in the season a game is played the less separation there is in that criterion.
Having said all that Miles biggest regular season win was 2005 at Alabama. Probably that Alabama team was not as good as this one, but they where 9-0 and also ranked #3. We were ranked #5 while we are #9 right now. That was a battle of top 5 teams, this one is not.

A regular season game win will never be bigger than winning a NCG, SECCG or a BCS bowl, period, end of story. Most bowl game will also be bigger wins.

I think it would definitely be this weekend. Miles' other REALLY big wins off the top of my head:
2005 AzSt - Simply due to the circumstances
2005 Miami
2006 Tennessee
2006 Arkansas
2006 Notre Dame
2007 VT
2007 UF
2007 Tennessee
2007 Ohio State
In all of these games, LSU was the better team. If we beat Bama, we will be beating the undefeated favorite on the road who has not lost an SEC game in almost two years. That, to me, is a bigger win than beating a less talented Ohio State team in virtually our own backyard.
Also, I do not think that Les Miles has beaten a team better than LSU during his tenure. A lot of that has to do with the fact that LSU is just better than most teams, but this game would definitely be the 1st.

Put it in this context:
If we beat Bama and go on to lose in the National Championship, will you think, "No big deal, that game against Bama was great?"
If we beat Bama and then lose another game and don't make a BCS bowl, will the Bama game be as big of a win?
If the answer to either is no, then it can't be the biggest win of the Miles era.

Beating Tennessee in SECCG and then beating Ohio State in the BCS title game will be hard to top.
But this would be a top 5 win for Miles...no doubt.

The biggest win was Fla. 2007 because without it, we are not in the BCS CG. The second is the BCS CG vs. Ohio State. Third is Va. Tech which was a total domination and also the huge margin is what vaulted LSU over Va. Tech and all the other teams who were ahead of us in that last week.

Also, you have to put Tennessee in there also, but all these wins were needed to be able to get the other win due to positions in the rankings.

The SEC CG is the bigget game any SEC school can play. Anything after that is lagniappe. The conference championship is what we all set out for. All else is circumstantial, ie.. Auburn going undefeated in the SEC and not playing for NC, or LSU loosing 2, and getting in NC because WVU lost. That is beyond conference teams control.
I believe the game against Alabama may be the biggest in Miles career.
In comparrisson, the 07 game against Fla. was huge in that it eatablished who was the team to beat in the SEC that year, and it established the kind of coach Miles was in how he coached with the team he had. He went for 5, 4th down conversions because he knew the personnel he had on the team and what they could do.
The 07 Tennessee SEC CG was big because it was..well...the SEC Championship.
The 08 BCS CG was big because it was for the championship. It kept LSU's, and the SECs reputations as a national power going, and established Miles as a prominent coach. Who really thought, going into that game that tOSU might win? Yea, the was some doubt early on, but really?
For me this weeks game is the biggest of Miles' career.
