The Soulsbourne boss that gave me the most deaths and frustration was Kalameet.
I know! Tons of people had no problem with that big dragon, but I could not get the timing right at all. I think it was because the DLC bosses were so much more aggressive than the main game. I had to relearn strategy. I struggled on all of them, but Kalameet destroyed me.
Technically speaking Nameless King, Orphan, Gael, etc. are more challenging bosses. Kalameet from DS1 was just brutal for me, though.
Edit: I should mention this was back when the DLC was released. Kalameet is easy to me now after playing these games for so long.

There are harder bosses in ninja gaiden, but I accidentally saved the game before the skeleton dinosaur with nothing to help me during the fight.
Had to play it perfectly with one bar of life. It took days before I had the perfect round.

The only boss isd argue that was harder in the regular mode only than Alma was spirit Doku. Those fish were an absolute nightmare to deal with while trying to kill him, especially unprepared.

he gave me headaches in the Mission Mode, but Lunar was usually able to make quick work of him. lots of RNG with that fight.

Bless those of you who have the masochistic tendencies to get through the DS games. Myself? No. Too much pain.
My personal toughest was the awakened form of Daimon in Dragon's Dogma Dark Arisen. I deliberately did not watch any videos or read reviews for spoilers.
I walked (ran) in there blindly at lvl 75 and barely beat the normal form before it "woke up". I spent the next 85 minutes battling him solo after he vortexed all my pawns, and went through 25 wakestones.
Of course after you beef up you can solo him in 15 seconds but I will never forget the frustration of the first fight. Still my favorite RPG ever

The Oryx Raid in Destiny's Taken King. Multiple first person jumping puzzles, four boss encounters and special challenge objectives. The entire raid takes hours to complete and required nearly flawless execution from all six players.. the phrase "that's a wipe" is PTSD inducing.

I hate bosses with obscenely large health pools, so Darkeater Midir comes to mind. Never solo'd him, didn't have the patience. I also summoned for Sinh, the Slumbering Dragon. frick dragons in the Souls games.
Oddly enough, none of the Sekiro bosses gave me too hard of a time (did the "good" ending). Being able to beat a lot of them via posture does wonders.
