Who you got?
My vote is Ted Knight as Judge Smails in Caddyshack! A majority of all the great moments of the movie were either delivered by him or in response to him yet he doesn't get near the recognition that Dangerfield, Murray, or Chevy received

Thomas Wilson in the BTTF series. He played young Biff, Old Biff, Middle Age wimpy Biff, Middle Age evil Biff, Griff, and Mad Dog Buford Tannen and killed it as each one.

Even though it is very short, Jon Hamm in Bridesmaids is very funny.
Also Jim Carrey in the The Incredible Burt Wonderstone.

Chris Tucker in The Fifth Element. He was the best part of a crazy movie. Well, him and Milla's outfit.

While I think of an answer to this thread, I'll have to use this as a place holder:
One of my favorite underrated lines is from Kyle Gas in Elf describing a children's book : we open on a farm with a group of asparagus children who are subconscious about the way their pee smells.
My vote for all time underrated comedy. Most people I talk to have never even heard of it. It has a great cast and plot and is a legitimately gold movie

Yeah, it's hard to beat Ted Knight in that movie. The underrated power player in the movie.
I'd go with Harold Ramis as Egon in Ghostbusters.

I'll have to go with Phillip Seymour Hoffman in Along Came Polly. The entire movie is worth watching just for his scenes. He was funny as hell playing those bagpipes, though.
Hon. Men.:
I'm tempted to say Daniel Stern as Brickma in Rookie of the Year.

Alan Rickman in Galaxy Quest. He simply made that film awesome.
"Miners not minors"
"Its like throwing gasoline on a flame"
"Could you possibly try not to hit every single one!"

I don't know about "in movie history" but my first thought was Michael Keaton as Billy Blazejowsky.
