I read on here that it could but I can't find that thread. Does anyone know if it works? Do I just walk up and hand them my TWIC card or do I need to do something else?

yes

go to your online reservation and type in the number on the lower left corner of the back of the card in the "known traveler" field. the TSAPre symbol will then print on your boarding pass

Yes!

You enter your TWIC card information when booking your flight (or go online and enter it later).
