Man I'm in the beating red heart of conservative rural Oklahoma and I've seen it. I've seen them pull over on the side of the road and pray to the East on several occasions. It's mostly longhaul drivers.

Show me where I said anything about "separation of church and state". You won't be able to, because I didn't.
Once again, show me in the Constitution where God, Jesus or Christianity are referenced. Something else you won't be able to do.
Show me in the Declaration of Independence where there is any reference to Jesus or Christianity. You won't be able to. There is one mention of "Nature's God" and one mention of a "Creator".
All of the Abrahamic religions - Judaism, Christianity and Islam - believe in a "Creator".
There is nothing in either of those two documents that references Christianity specifically.
"Nature's God" sounds almost Wiccan. The Wiccan's have a better case to make than Christians.
The bottom line is that the First Amendment, which is codified law, prohibits Congress from prohibiting the free exercise of any religion, including Islam.

In my opinion, yes, as long as that business is in no way financially dependent upon any government entity.
However, all legitimate businesses are financially dependent on multiple government entities (tax law). That is how they get you. If you want to operate a legitimate business within the United States, then you're probably not going to be able to ban Muslim employees or customers from praying on your premises.

Ummm, maybe you should read the entire sentence where Jefferson specifically links text from the First Amendment directly to "separation of church and state".
Here it is:
