Wayne didn't have cancer when it was filmed, but it perfectly tells his story.

The Duke is who we all should emulate when smoking cigs. Only used one match a day for his first cig, lit the rest of his 6 packs a day with the previous cig.

A few other nominees:
Randolph Scott, Ride The High Country -- it was going to be Joel McCrea's too, but he made a couple of appearances 10-15 years later
Edward G. Robinson, Soylent Green

Edward G. Robinson, in Soylent Green is my pick.
Watch the scene were he is dying, and realize he dead that day or a few days later.
Edward filmed the movie knowing he only had a little time left.
Edward G. Robinson is the most underrated actor or one of the most underrated, along with Paul Muni.

Bela Lugosi in Plan 9 From Outer Space.

A few other opinions:
James Dean -Giant
Clark Gable -The Misfits
Oliver Reed. -Gladiator
Would like to make a case for Peter Finch in Network, but I thought the film overrated.

Another contender is Spencer Tracy, who was nominated for an Oscar for Guess Who's Coming to Dinner, and passed away just days after production ended.

and FWIW I also think Network is overrated (and Giant too, aside from Dean's early scenes, for that matter; I've never seen Gladiator)

The Last Movie Star - Burt Reynolds

He was pretty ill, a lung had been removed, heart issues.
“The actor had had a lung removed twelve years earlier and was now struggling with the stomach cancer that would eventually lead to his death in 1979.”

Let's set aside our differences about the film and focus on, particularly, the key performances - Dunaway, Finch and Holden - we can agree they all hit home runs can we not?
And the supporting cast was amazing - Straight, Duvall and Beatty gave outstanding performances (although not unusual for that group, either).

It's not great history, but it is great film.

I hope somebody somewhere is writing at least one more great western that ends up being Duvall’s final movie.
Hopefully he has years of movies left, but I hope he goes out with a great western.

He's literally 90 - the same age as Eastwood and Shatner. That great shot clock in the sky runs against every single one of us.

Wayne's death led to my parents meeting for the first time.
My Dad had lowered the flag to half-mast atop a Famous Barr where he worked that day, got in trouble because of it and sent to the Managers/HR office where my Mom was the assistant. Asked her if she wanted to go to a concert the next day.

I was gonna say Heath Ledger in The Dark Knight, but I overlooked his actual last film, The Imagination of Dr. Parnassus, which I had never even heard of. Had a pretty good cast too.

Burgess Meredith in Grumpier Old Men.
Got laid, got to sing a song to his granddaughter, and his last scene was The Bacon Story.
Can’t end your career on a higher note than that.

I believe he died during the production.
He was pretty out of control most of his career until the very end and was always up for a drink.
