Pacino in Scent of a Woman.
I also enjoy Dustin Hoffman in Hook.

Lots of good names in this thread. Of those listed I'd go with Duval in Lonesome Dove (though he was also great in a range of other movies) and Brando in The Godfather.
My dark horse: Boggie in The African Queen. He played against type for his usual roles and I thought he did a great job as the scraggly rundown boat captain falling slowly in love with Katherine Hepburn.

I really have never understood the love for Marlon Brando in the Godfather. What does he do that's that amazing, or even moderately impressive, at all? It's a fat guy mumbling his lines. Who cares? I don't get the love for the Godfather in general. I thought I & II were both boring. Give me Goodfellas over the Godfather any day.
Now, Charlize in Monster, Daniel Day Lewis in anything he's done, Leo in Gilbert Grape - that's acting.
